Staining
Hematoxylin-Eosin is a commonly used tissue stain. Hematoxylin stains nucli blue and eosin stains the extracellular matrix and cytoplasm pink. This techniques uses hazardous Xylene as a deparaffinizing agent. Exposure also occurs during tissue processing, placing the coverslip and cleaning.
Try replacing it with dish washing solution or other non-hazardous alternatives.
Prema et al. showed that all three - 17% dishwashing soap, a 95% lemon water and a 100% coconut oil - could be used as a safer and cost-effective substitute for xylene. All gave good results.
Sravya et al. processed tissue with sesame oil stained with 1.7% dish washing solution as an alternative to xylene.
Pandey et al. also reported dishwashing solution as a great alternative to xylene.
Premalatha et al. used mineral oil as a substute for xylene.
