Branch of scinece that studies microscopic anatomy of biological tissues.

# Staining

Hematoxylin-Eosin is a commonly used tissue stain. Hematoxylin stains nucli blue and eosin stains the extracellular matrix and cytoplasm pink. This techniques uses hazardous Xylene as a deparaffinizing agent. Exposure also occurs during tissue processing, placing the coverslip and cleaning.

Try replacing it with dish washing solution or other non-hazardous alternatives.

[Prema et al.](https://www.ncbi.nlm.nih.gov/pmc/articles/PMC7595545/) showed that all three - 17% dishwashing soap, a 95% lemon water and a 100% coconut oil - could be used as a safer and cost-effective substitute for xylene. All gave good results.

[Sravya et al.](https://www.ncbi.nlm.nih.gov/pmc/articles/PMC5917531/) processed tissue with sesame oil stained with 1.7% dish washing solution as an alternative to xylene.

[Pandey et al.](https://pubmed.ncbi.nlm.nih.gov/25328332/) also reported dishwashing solution as a great alternative to xylene.

[Premalatha et al](https://pubmed.ncbi.nlm.nih.gov/23811660/). used mineral oil as a substute for xylene.
